I didn't learn this until after I was stable at my optimal levels.
I do know that it's best to be consistent with the time of day you get your blood drawn....that way, you'll have a clear picture moving forward.
Hopefully, your doctor is monitoring your FreeT4 and FreeT3 levels.
Some people make sure to not take their levo for at least 8 hours before the draw - there doesn't seem to be any consistency with my research as to whether or not this is appropriate.
I take my levo at night so, this isn't an issue for me.

I would say it's best to take your level at least 8 hours before the draw or wait until after the draw to take it.
25mcg is a common starting dose and is rarely a therapeutic dose.
You will most likely need many dose increases to restore wellness.
That said, I wouldn't worry about the day 3 of the cycle idea.
It's more important to get labs every 6 wks. (the amount of time it takes for things to stabilize after a dose adjustment).
It just doesn't make sense to me to miss an opportunity for a needed dose adjustment if lab timing doesn't work well with our cycles.
