We backcrossed the IL2Rγnull allele onto NOD-scid mice, a commonly used mouse strain for human HSC engraftment . Before testing human HSC engraftment in NOD-scid IL2Rγnull mice, we first defined the phenotypic, histological, and functional properties of their lymphohemopoietic system. DNA was isolated from single-cell suspensions from spleen and thymus of engrafted mice using the QIAamp DNA mini Kit following the manufacturer’s instructions. RNA was purified from the same samples with the RNeasy Mini Kit following the manufacturer’s instructions. As they are made of polymer, they are opaque making it difficult to see where each sample should go when transferring many.

FITC-conjugated anti-CD86 mAb , PE-conjugated anti-CD11c mAb , PerCP-conjugated anti-CD11b mAb (clone M1/70), and AP-conjugated anti-TNF-α (clone MP6-XT22) were obtained from BD Biosciences.
We report the development and characterization of a new genetic stock of IL-2R common γ-chain deficient NOD/LtSz-scid (NOD-scid IL2Rγnull) mice and document their ability to support human mobilized blood HSC engraftment and multilineage differentiation. NOD-scid IL2Rγnull mice are deficient in mature lymphocytes and NK cells, survive beyond 16 mo of age, and even after sublethal irradiation resist lymphoma development. Engraftment of NOD-scid IL2Rγnull mice with human HSC generate 6-fold higher percentages of human CD45+ cells in host bone marrow than with similarly treated NOD-scid mice. These human cells include B cells, NK cells, myeloid cells, plasmacytoid dendritic cells, and HSC. Spleens from engrafted NOD-scid IL2Rγnull mice contain human Ig+ B cells and lower numbers of human CD3+ T cells. Coadministration of human Fc-IL7 fusion protein results in high percentages of human CD4+CD8+ thymocytes as well human CD4+CD8− and CD4−CD8+ peripheral blood and splenic T cells.

The accuracy determined at quality control levels was within 92.81–105.13%. These results show that Mam-A2.2–specific CD8 + CTLs are normally expanded in humans. This recognition may be due to cross-reaction between the Mam-A2.2 peptide and a naturally presented peptide derived from a cross-reacting environmental pathogen recognized by the T cells of HLA-A2 + individuals. However, the low-level expression of mammaglobin-A in normal breast tissue might also lead to the development of autoreactive T cells toward mammaglobin-A–expressing breast tissue.
This could be due to a lower level of antibody which is not detected by the MAT at a dilution of 1 in 50 or a non-specific antibody reaction. In this study, 41% (74/180) of the leptospirosis IgM ELISA reactive samples had reactive IgG and/or IgM serology on the MIA, again suggesting the level of antibody in these particular samples may be too low for the MAT to detect. Also, this again shows that there may be some non-specific reactions occurring in the IgM ELISA, which are not seen on the MIA. The MIA is therefore advantageous as a screening test as it reduces the large numbers of samples that are unnecessarily sent for confirmation testing by MAT.

This constrained our ability to conduct determinations of total cellularity of lymphoid organs. Therefore, we did not measure the effect of Fc-IL7 treatment on total numbers of T and B cells. Future quantitative studies investigating the dose and duration of IL-7 on human T and B cell development in these mice are under way. Ethical considerations constrain the in vivo study of human hemopoietic stem cells . To overcome this limitation, small animal models of human HSC engraftment have been used.
